Synthesis of acetoacetate by liver enzymes.

چکیده
Two mechanisms have been proposed for the formation of acetoacetate from acetyl-CoA by liver preparations. With the use of a partially purified system prepared from acetone powders of beef liver, Lynen et al. (1) obtained evidence that acetoacetate formation proceeded via P-hydroxy-fi-methylglutaryl coensyme A as intermediate and involved the HlMG-COAT condensing and HMG-Coh cleavage enzymes as shown in Reactions 1 and 2.

On the mechanism of acetoacetate synthesis by guinea pig liver fractions.
1. Commercially available coenzyme A preparations were found to contain sufficient amounts of glutathione so that in the reaction with diketene significant quanties of acetoacetyl glutathione were formed. Unless allowances are made for this, significant errors in the interpretation of results can be made because liver mitochondria contain a very active acetoacetyl glutathione hydrolase. 2. Puri...
متن کاملThe mechanism of acetoacetate synthesis.
The observation of a coenayme A (CoA)-catalyzed acetoacetate synthesis from acetate and adenosinetriphosphate in pigeon liver extracts (1) grouped this reaction with the larger family of acetylation reactions. Furthermore it furnished partial confirmation of an earlier calculation (2) showing that 16,000 calories are required for the condensation of two acetates to acetoacetate. Thus, one energ...

The synthesis of citrate from acetate, ATP,’ and oxalacetate by soluble enzyme preparations from pigeon liver was briefly reported in 1949 (1). CoA was found to be required for activity. Similar results were later reported by Novelli and Lipmann (2) with preparations from yeast and Escherichiu coli, and by Elliott and Kalnitsky (3) with kidney acetone powder extracts. It was further shown (1) t...
متن کاملStudies in the ketosis of fasting.
A series of experiments was performed during the induction of starvation ketosis and in the acute reversal of the ketotic state. In contrast to the predictions of two widely held theories of ketogenesis, control of acetoacetate production by the liver appeared to be unrelated to changes in fatty acid mobilization from the periphery, fatty acid oxidation, fatty acid synthesis, or the acetyl coen...
متن کاملStudies of fatty acid oxidation. 3. The effects of acyl-CoA complexes on fatty acid oxidation.
The oxidation of fatty acids to acetoacetate or to carbon dioxide is affected by the presence of various aliphatic and aromatic acids. For example, benzoate inhibits acetoacetate formation from butyrate at low concentrations in guinea pig liver slices (Jowett & Quastel, 1935b) and in 'cyclophorase' preparations (Grafflin & Green, 1948). Benzoate also inhibits the oxidation of butyrate to C02 in...
